Product overview

The moisture-proof excipient with a cooling edge

Mannitol (D-mannitol) is a six-carbon sugar alcohol found naturally in seaweed, mushrooms and the manna ash tree. Industrially it is produced by catalytic hydrogenation of fructose or glucose-fructose syrups, yielding a white, odourless crystalline powder with approximately 50 % the sweetness of sucrose.

In pharmaceutical formulation, mannitol's stand-out property is its non-hygroscopic nature — it does not absorb atmospheric moisture even at elevated relative humidity. This makes it the excipient of choice for moisture-sensitive APIs that would degrade on contact with water. Its endothermic heat of dissolution (negative ΔH) produces a pleasant cooling sensation in the mouth, making it the standard sweetener-filler in chewable and orally disintegrating tablets (ODTs).

For lyophilised (freeze-dried) parenterals, mannitol crystallises during the freezing step to form a mechanically rigid cake that supports rapid reconstitution with minimal shrinkage. It is the single most common bulking agent in injectable lyophilisates, used alongside amorphous cryoprotectants such as sucrose or trehalose.

Beyond excipient use, mannitol itself is a pharmacologically active substance: intravenous mannitol 20 % is a first-line osmotic diuretic for managing raised intracranial pressure, cerebral oedema and acute oliguric renal failure. Inhaled dry-powder mannitol is also used as a diagnostic agent in the bronchial provocation (mannitol challenge) test for airway hyper-responsiveness in asthma.

Applications & end uses

Industries that depend on Mannitol

Non-hygroscopic tablet excipient

Mannitol is the excipient of choice for moisture-sensitive APIs. Unlike lactose and sorbitol, mannitol does not absorb atmospheric moisture, ensuring tablet stability and consistent disintegration even in tropical and high-humidity climates.

Chewable tablet sweetener

The endothermic dissolution of mannitol produces a pleasant cooling sensation in the mouth, combined with approximately 50 % of the sweetness of sucrose. This makes it the preferred sweetener-diluent in chewable and orally disintegrating tablets.

Lyophilisation bulking agent

Mannitol crystallises during freeze-drying to produce an elegant, rigid cake with rapid reconstitution. It is the most commonly used bulking agent in lyophilised parenterals, providing mechanical structure and protecting the API during the sublimation cycle.

Osmotic diuretic (IV mannitol 20 %)

Injection-grade mannitol administered intravenously as a 20 % solution is a first-line osmotic diuretic for the management of raised intracranial pressure, cerebral oedema, acute renal failure and glaucoma in hospital settings.

Sugar-free confectionery

Mannitol is widely used in sugar-free chewing gum, mints, chocolate coatings and hard-boiled sweets. Its low caloric value (1.6 kcal/g versus 4 kcal/g for sucrose), non-cariogenic nature and cooling effect make it ideal for diabetic and dental-health products.

Mannitol challenge test (asthma diagnosis)

Inhaled dry-powder mannitol is used as a diagnostic agent in the bronchial provocation (mannitol challenge) test for assessing airway hyper-responsiveness in asthma, approved as Aridol / Osmohale in multiple markets.

Frequently asked questions

Common questions about Mannitol

Why is Mannitol preferred over Lactose for moisture-sensitive APIs?+
Mannitol is non-hygroscopic — it does not absorb atmospheric moisture even at high relative humidity. Lactose monohydrate, by contrast, contains crystal water and can interact with moisture-sensitive compounds. This makes mannitol the excipient of choice for APIs that are prone to hydrolysis or degradation on exposure to moisture.
Which pharmacopoeia grades of Mannitol does Chelora supply?+
We supply mannitol meeting IP, BP and USP monograph specifications, including pharma-grade powder, spray-dried/granular (DC) grades and injection-grade (pyrogen-free) mannitol for parenteral applications. Each batch includes a COA confirming compliance.
Is Mannitol safe for diabetic patients?+
Mannitol is a sugar alcohol with a low glycaemic index and caloric value of approximately 1.6 kcal/g (vs 4 kcal/g for sucrose). It is widely used in sugar-free formulations suitable for diabetic patients. However, excessive oral intake may have a laxative effect, as noted in pharmacopoeia monographs.
What gives Mannitol its cooling sensation in chewable tablets?+
Mannitol dissolves endothermically — it absorbs heat from the surrounding environment (the mouth) as it dissolves, producing a distinct cooling sensation. This property, combined with its moderate sweetness, makes mannitol the standard sweetener-diluent for chewable and orally disintegrating tablets.
What is the typical lead time for a bulk order?+
Standard lead time is 5 – 10 working days for domestic orders and 10 – 14 days for export, depending on grade and volume. Injection-grade mannitol may require slightly longer lead times due to additional quality testing.
Do you offer samples before bulk purchase?+
Yes. Sample quantities of all three grades are available so your QC and R&D teams can verify the material against your specification before committing to a production order.
