Product overview

Rapid swelling for rapid release

Croscarmellose sodium is a cross-linked polymer of carboxymethylcellulose sodium, engineered to swell rapidly on contact with water while remaining insoluble. This controlled insolubility is the key to its function as a super-disintegrant: the polymer absorbs water, swells to many times its dry volume and generates the internal pressure that breaks the tablet matrix apart within seconds.

Manufactured by cross-linking sodium carboxymethylcellulose with subsequent acid washing and drying, the resulting fibrous white powder retains a high degree of substitution (0.60–0.85) that drives the swelling mechanism. Unlike sodium starch glycolate, croscarmellose sodium does not form a viscous gel layer at typical use levels, which avoids any retardation of drug release.

It is effective at low concentrations — typically 2–5 % w/w — and can be incorporated both intragranularly and extragranularly for a dual disintegration effect. This versatility, combined with compatibility with most APIs, makes it one of the three super-disintegrants listed in every major pharmacopoeia (IP, BP/Ph. Eur., USP-NF).

Applications extend beyond standard tablets to orally disintegrating tablets (ODTs), capsule fills, granules and co-processed excipient systems. In each case, croscarmellose sodium ensures that the dosage form breaks down quickly enough for the API to dissolve and be absorbed within the therapeutic window.

Applications & end uses

Where Croscarmellose Sodium delivers performance

Tablet super-disintegrant (primary use)

Croscarmellose sodium is one of the three major super-disintegrants used in immediate-release tablets. At 2–5 % w/w it provides rapid disintegration by swelling on contact with gastrointestinal fluid, breaking the tablet matrix into primary particles for fast dissolution.

Capsule disintegrant

Incorporated in hard gelatin and HPMC capsule fills, croscarmellose sodium promotes rapid deaggregation of the encapsulated powder plug, ensuring uniform drug release equivalent to tablet formulations.

Orally disintegrating tablets (ODTs)

ODT formulations rely on super-disintegrants to achieve disintegration within seconds in the oral cavity without water. Croscarmellose sodium’s rapid swelling and wicking action make it a preferred choice for fast-melt and lyophilised ODT platforms.

Granule disintegrant

When added intragranularly (within the granule) and/or extragranularly (outside the granule), croscarmellose sodium provides a dual disintegration mechanism — first breaking granules into fine particles, then facilitating complete deaggregation.

Co-processed with MCC for direct compression

Croscarmellose sodium is often co-processed with microcrystalline cellulose in direct-compression platforms. The combination delivers good compactibility with built-in disintegration, simplifying formulation and reducing manufacturing steps.

Nutraceutical formulations

Dietary supplement tablets and capsules use croscarmellose sodium to ensure rapid disintegration and dissolution of vitamins, minerals and herbal extracts, meeting USP <2040> dissolution requirements for nutritional products.

Frequently asked questions

Common questions about Croscarmellose Sodium

What is the typical use level of Croscarmellose Sodium in tablets?+
The typical concentration is 2 – 5 % w/w of the tablet weight. For most immediate-release formulations, 2 – 3 % is sufficient. Higher levels (up to 5 %) may be used for formulations with hydrophobic APIs or high drug loading where faster disintegration is needed.
How does Croscarmellose Sodium compare with other super-disintegrants?+
Croscarmellose sodium, sodium starch glycolate (SSG) and crospovidone are the three major super-disintegrants. Croscarmellose sodium works primarily by swelling and wicking, SSG by swelling, and crospovidone by wicking and strain recovery. Croscarmellose sodium is often preferred because it does not form a gel at high concentrations, unlike SSG.
Should Croscarmellose Sodium be added intragranularly or extragranularly?+
For optimal disintegration, a split addition — part intragranularly (within the granule) and part extragranularly (blended after granulation) — is recommended. This provides two stages of disintegration: granule break-up followed by fine-particle deaggregation. A 50:50 or 25:75 (intra:extra) split is common.
Is Croscarmellose Sodium suitable for moisture-sensitive formulations?+
Croscarmellose sodium absorbs moisture and can accelerate degradation of moisture-sensitive APIs if the formulation is not appropriately packaged. For such products, moisture-barrier blister packs or desiccant-containing containers are recommended. Crospovidone may be preferred in extreme cases.
Is Croscarmellose Sodium classified as a hazardous material?+
No. Croscarmellose sodium is not classified as dangerous goods for road, sea or air transport. Standard packaging and labelling apply. An SDS is supplied with every shipment for your EHS records.
What is the typical lead time for a bulk order?+
Standard lead time is 5 – 10 working days for domestic orders and 10 – 14 days for export shipments, depending on grade and volume. We can often accommodate expedited dispatch for urgent production schedules.
